Abstract

Method’s validatio is very important in research, including in the antioxidant activity test. Based on USP XXXVII 2014, antioxidant activity test is included in category III research where analytical methods are used to ensure the performance and quality of finished product. Precision is a must in this cathegory and other parameters may be needed. This research aims to test the validation of the linearity of the antioxidant activity method of ascorbic acid using DPPH with ethanol as the antioxidant testing solvent. This study used ascorbic acid samples with 96% p.a. ethanol solvent. The test was conducted by taking 10 mg of ascorbic acid with concentrations of 1, 2, 3, 4, 5 ppm and repeating it 3 times. The data obtained concluded that the absorbance results from replicates 1, 2, and 3 were not significantly different. As the concentration increased, the resulting absorbance decreased. The percentage decrease results from this research can be considered valid because the obtained r values are close to 1, namely replication 1 = 0.9932, replication 2 = 0.9946, replication 3 = 0.9910.
