Abstract

Diabetes mellitus in Indonesia has increased to 8.5% in 2018. To overcome this can be done by inhibiting the ?-amylase enzyme so that it can reduce blood glucose. One herbal plant that is effective in lowering blood sugar levels is black ear mushroom (Auricularia polytricha). Black ear mushroom contains flavonoids that have the potential to reduce blood glucose levels. The purpose of this study was to determine the antidiabetic potential of black ear mushroom fraction using different solvents in inhibiting ?-amylase enzyme. Black ear mushroom was extracted using maceration method with methanol solvent and then liquid-liquid fractionation. The water, chloroform and ethyl acetate fractions of black ear fungus obtained were subjected to phytochemical screening and ?-amylase enzyme testing in vitro using dinitrosalicylate (DNS) reagent. Inhibition of ?-amylase enzyme was carried out at concentrations of 50; 100; 200 400; 600; 800 and 1000 ppm. Data analysis was done descriptively by calculating % inhibition and IC50 value. The results showed that there were flavonoids in the water fraction; steroids in the chloroform fraction; and flavonoids in the ethyl acetate fraction. The IC50 value of the ethyl acetate fraction was 99,606 ?g/mL in the strong category and water was 99,714 ?g/mL in the strong category, while the chloroform fraction was 166,398 ?g/mL in the weak category. The ethyl acetate fraction showed the highest inhibition compared to other fractions.
